Lupus is an auto immune disorder in which the patient's body generates antibodies against its own organs and tissues. IgG antibodies generated against some of the body's own tissues, interact with white blood cells. This causes the nucleus of white blood cells to degenerate. This degenerated nucleus, along with portions of the white cells, are ingested by phagocytes which results in the formation of LE cells. Phagocytes are part of every person’s immune system which help in annihilating any potential threat to human body. Formation of LE cells is a pathological indication of underlying auto immune disorders.

Positive findings of LE test indicate presence of Lupus and negative findings rule out the condition completely. A venous blood sample is collected for the test. Labs offering histology services are equipped to perform this test. A smear is prepared out of the obtained blood sample which is later studied under a high resolution microscope.

To prepare the smear, a tiny amount of the patient’s blood sample is required. Blood is placed at one end of a microbiology slide. Using another slide the blood is smeared till it forms an even coat over the first slide. While preparing a smear, care needs to be taken to avoid formation of air bubbles. The smear is allowed to dry well before it is studied. The slide is retained in extremely sterile environment till the time it is studied.

A high resolution microscope is used to study the prepared slide. Finding over 10 LE cells over a span of 15 minutes of studying the smear is considered to be a positive finding. The speed with which the slide is to be moved during these 15 minutes is pre designated. Alternatively, the same procedure may be performed using a bone marrow sample. However, since this could be painful to the patient, bone marrow is used very rarely.

Other than Lupus, LE cells may also be detected in blood samples of patients suffering from disorders like rheumatoid arthritis, polyarteritis nodosa, dermatomyositis. Patients who are prescribed LE cells test should consult with a qualified hematologists before undergoing the procedure.
